沙柳组织培养的初步研究

摘    要:以沙柳叶片和茎段为外植体,采用WPM、MS、F14为基本培养基,附加6-BA与NAA 2种激素组合,摸索了适合沙柳愈伤组织诱导的基本培养基、最佳激素组合浓度以及最适合的外植体。研究结果表明:WPM为最适合愈伤诱导的基本培养基,最佳外植体是茎段,最佳激素组合浓度为6-BA 0.5 mg/L+NAA 1.5 mg/L,诱导率达87.5%。本研究开展了沙柳组织培养的初步研究工作,这对于沙柳优良无性系的组培快繁体系建立、沙柳细胞遗传学的研究以及基因转化受体系统建立等奠定了研究基础。

Preliminary Research on Tissue Culture of Salix psammophila

Abstract:Taking the leaves and stem pieces of Salix psammophila as the explant, the WPM, MS and F14 as mini- mal medium, with the hormones of 6 -BA and NAA, the tissue culture researches have been conducted. The re- sults show that WPM is the minimal medium most suitable for callus of induce, and the suitable explant is stem pieces. The hormones combination and concentration are 6 - BA 0.5 mg/L + NAA1.5 mg/L with the highest callus induction frequency of 87.5 %. This research has laid a foundation for fine clonal propagation of Salix psammophila by tissue culture, cytogenetics research work and gene transformation by plantlet regeneration system.
