小麦淀粉GBSSII基因的克隆、反义和RNAi载体的构建

摘    要:采用RT-PCR方法从小麦品种豫教2号籽粒中克隆出颗粒型淀粉合成酶II基因(Granule bound starch syn-thase,GBSSII)部分cDNA片段(1 069 bp)(GenBank登录号:EF221764),同源性比较结果显示,它与GenBank上已报道的GBSSII基因有高度的同源性。以pWM101质粒为基础,构建了由35 S启动子调控的GBSSII基因的反义表达载体pWM101GBSSIIA;另外,还以pFGC5941质粒为基础,构建了GBSSII基因的RNAi载体pFGC5941GBSSIIsa,这些载体的构建为研究GBSSII基因的功能打下了很好的基础。

Abstract:Partial cDNA sequences(1 069 bp)of granule bound starch synthase II(GBSSII)gene(GenBank No.EF221761)was amplified from grains of common wheat cultuvar Yujiao 2 by RT-PCR in the paper.The result demonstrated that the cloned GBSSII gene sequence had 95%-97% identity with the reported GBSSII genes in GenBank.In addition,its antisense expression(pWM101GBSSIIA)vector was constructed with pMW101 vector,and RNAi vector(pFGC5941GBSSIIsa)was also constructed with pFGC5941.These constructed vectors would provide a good background for investigting the function of GBSSII on biosynthesis of starch in wheat plants.
