西藏青稞育成品种醇溶蛋白的遗传多样性分析

摘    要:利用A-PAGE(Acid-polyacrylamide gel electrophoresis)法对18份西藏青稞育成品种醇溶蛋白的遗传多样性进行了研究。结果表明:18份供试材料共分离出22种相对迁移率不同的醇溶蛋白谱带,16种不同的电泳图谱,说明西藏青稞育成品种具有较丰富的醇溶蛋白遗传多态性。平均等位变异数为10个,平均遗传多样性值为2.602。聚类分析结果表明,四川农业大学育成的青稞品种2000-9与所有的西藏材料分别聚为A、B两大类。在遗传距离为0.42水平上,所有的西藏材料又可归为B1、B2两亚类。遗传距离分析表明,西藏材料间的遗传距离平均值较低(0.282)。在西藏超级青稞育种中,应加强青稞资源的收集、发掘和评价,扩大青稞资源的遗传基础。

Abstract:Genetic diversity of hordein in 18 accessions of hulless barley varieties bred in Tibet was analyzed by acid-polyacrylamide gel electrophoresis(A-PAGE) in this paper.The results showed that a total of 22 hordein bands with relatively different mobilities and 16 types of hordein patterns were identified in these accessions from Tibet,which indicated that richer genetic polymorphism of hordein was existed in the hulless barley varieties bred in Tibet.The minimum number of allele is 10,and the average genetic diversity is 2.602.The cluster analysis revealed that,the hulless barley variety "2000-9" bred by Sichuan Agricultural University and all the accessions from Tibet were separately clustered into two major groups: A and B.At the genetic distance level of 0.42,all the Tibetan entries in Group B were classified into two subgroups : B1 and B2.And yet lower mean genetic distance(0.282) among these materials from Tibet was estimated.Finally,it suggested that the collection,excavation and evaluation of hulless barley resources should be strengthened in order to widen genetic basis of hulless barley resources in over-hulless barley breeding in Tibet.
