| 摘 要： | 半套式 PCR是采用一对半特异性引物对目的基因进行扩增的一种方法 ,该方法较常规 PCR的更敏感和更特异。本试验即采用该种方法 ,从石蜡包埋组织中提取圆环病毒 DNA进行 PCR扩增 ,来检测猪圆环病毒。结果表明组织石蜡切片经二甲苯脱蜡 ,蛋白酶 K消化 ,利用半套式 PCR方法 ,得到 43 2 bp的特异性扩增产物 ,从而建立了一种新的检测圆环病毒的方法 ,并为从存档石蜡研究该病提供了很好的前景

| Abstract: | Two serial specific primers(P1/P2,P2/P3) were used in semi nested PCR,to amplify predicated gene,which is more sensitive and specific than conventional PCR.In this study,semi nested PCR was developed for the detection of porcine circovirus 2(PCV2) from formalin fixed paraffin embedded tissues.The result indicated,after xylene deparaffinization followed by proteinase K digestion, the predicated products,432 bp specific fragement, were amplified successfully from paraffin sections by semi nested PCR.So a new method was established,which gave a good chance to study PCV2 from archival paraffin embedded tissues. |
