摘    要:对适用于玉米的RAPD实验体系进行研究,摸索出适用于玉米的RAPD的反应适宜条件闻稳定的玉米RAPD分析体系,应用这一体系对温敏核雄性不育玉米琼42-Qms及其可育的近等基因系琼42进行RAPD分析,在300多个引物中发现1个引物在两种中扩增带型有差异,经重复试验,初步认为此差异与玉米雄性不育基因连锁。

Abstract:Various experimental protocals for RAPD analysis of genes in maize were systematically studied and an optimum program was developed for setting up of a stable RAPD analysis system for maize. By using this system,thermal -sensitive genie male sterile maize Qiong42 -QMS and its near -isogenic line Qiong42 were analyzed. More than 300 primers were used for RAPD amplification of Qiong 42-QMS and its near-isogenic line Qiong42,and a polymorphic DNA fragment about 520 hp was obtained with only one primer No. 320. This fragment was only found in the sterile maize and its lines but not in the non -sterile maize lines. Repeated experiments proved this fragment had a good repeatability,and it was preliminarily believed to be linked to the maize thermal -sensitive male sterility gene.
