Abstract:In vivo antitumor activity of ergosterol from fruiting bodies of Agaricus blazei Murrill was tested by using mouse S180 transplant tumor model. The antitumor mechanism of ergosterol were preliminary studied by a series of tests such as phagocytosis of macrophagus, proliferation of splenic cells, cytotoxicity and angiogenesis of chick chorioallantoic membrane (CAM). The tumor weight inhibition rate of ergosterol from fruiting bodies of Agaricus blazei Murrill reached 62.7%, 76.6% and 79.1% at doses of 10 mg·kg^-1d^-1, 50 mg'kg^-1 ·d^-1 and 100 mg·kg^-1d^-1respectively in vivo test, showing strong antitumor activity of ergosterol. However, ergosterol did not show immunologic enhancement in tests of phagocytosis of peritoneal macrophages and proliferation of splenic lymphocytes. Additionally, ergosterol showed no evident cytotoxicity against tumor cells such as SGC-7901 and Bel-7402 in vitro test. The CAM assay showed that ergosterol had stronger anti-angiogenetic effect (inhibition rate 63.46% and 78.78% at doses of 5 μg·mL^-1 and 50 μg·mL^-1 respectively) as compared to dexamethasone. It seems likely that the antitumor activity of ergosterol might be due to direct inhibition of angiogenesis induced by tumors.
