细叶百合DREB转录因子生物信息学及胁迫应答表达分析

摘    要:DREB转录因子是AP2/ERF家族的主要成员,在植物应对非生物胁迫中发挥重要作用。目前还没有针对细叶百合DREB转录因子家族的系统分析。本研究基于细叶百合(根、茎、叶)转录组数据筛选出12个DREB转录因子,命名为LpDREB1-LpDREB12,并对其进行生物信息学及不同组织在胁迫下的表达模式分析。结果表明,细叶百合DREB蛋白多为不稳定蛋白且具有一定亲水性,α螺旋和无规则卷曲是蛋白二级结构的主要组成部分。系统进化分析显示,12个细叶百合DREB转录因子同61个拟南芥DREB转录因子聚类到一起,说明细叶百合和拟南芥DREB家族成员具有较高的保守性。表达模式分析表明,在干旱、盐、低温及脱落酸胁迫下12个细叶百合DREB基因的表达均有组织特异性。研究结果丰富了细叶百合DREB基因数据库,为挑选优质抗逆基因提供了理论依据。

Abstract:DREB transcription factor is a major member of AP2/ERF family and plays an important role in plant response to abiotic stress.At present,there is no systematic analysis of DREB transcription factor family in Lilium pumilum.Based on the transcriptome data of L.pumilum(root,stem and leaf),12 DREB transcription factors were selected and named LpDREB1 to LpDREB12,and their bioinformatics and expression patterns in different tissues under stress were analyzed.The results showed that most of the DREB proteins of L.pumilum were unstable proteins and had certain hydrophilicity,α-helix and irregular curl were the main components in protein secondary structure.Phylogenetic analysis showed that 12 DREB transcription factors of L.pumilum and 61 DREB transcription factors of Arabidopsis thaliana clustered together,indicating that L.pumilum and DREB family members of A.thaliana had high conservatism.The expression pattern analysis showed that the expression of 12 DREB genes in L.pumilum under drought,salt,low temperature and ABA stresses were tissue-specific.The results of this study enriched the DREB gene database of L.pumilum,and provided a theoretical basis for selecting high-quality stress resistance genes.
