一种新的蒺藜苜蓿遗传杂交方法

摘    要:遗传杂交技术是开展遗传研究的基本工具,也是研究基因功能的必要手段之一。虽然对豆科模式植物蒺藜苜蓿(Medicago truncatula)的研究愈来愈多,但其遗传杂交技术因难度较高而只被研究蒺藜苜蓿比较悠久的科研团体掌握。为了便于更多实验室能够掌握该项技术,本文在已报道的蒺藜苜蓿杂交方法的基础上提出了一种新的杂交技术。研究内容包括蒺藜苜蓿的花果发育周期、花解剖结构、不同发育时期花的花药发育、遗传杂交程序、母本花的选择标准以及杂交鉴定等。本研究所提出的蒺藜苜蓿杂交方法与已有其它杂交方法相比,具有操作难度低、杂交后无需特殊护理措施、杂交成功率高等特点。

Abstract:The technique of genetic crossing is a basic tool for genetic research and one of the necessary means to study gene function. Although there are more and more studies on the legume model plant Medicago truncatula, due to the difficulty of genetic hybridization of this material, its hybridization technique is still limited to the scientific research groups having utilized this plant material for a long time. In order to facilitate more laboratories to master this technology, this study proposed a new hybridization technique based on the reported hybridization methods of M. truncatula. The study included the flower and fruit development cycle, anatomical structure of flower, anther development of flowers at different stages, genetic hybridization procedures, selection criteria of female parent flowers, and identification on the successful hybridization. Compared with other existing methods of M. truncatula hybridization, the method proposed in this study featured some advantages, such as low difficulties on operation, no need for special nursing cares after hybridization, and high rate of successful hybridization.
