黑穗醋栗试管苗生产工艺流程的研究

摘    要:采用黑穗醋栗“丰产薄皮”与“亮叶厚皮”品种,对茎尖组织进行离体培养。通过调整培养基中BAP含量,获得了每20d达4.7的幼苗分化指数,幼苗在半量的MS无激素培养基上暗处理1~3d,接种密度每瓶3苗,生根率达83.3%~96.0%;将生根苗移栽于装有蛭石的木箱中,成活率达97%,并成功地移栽于大田,试管苗生长旺盛。建立了黑穗醋栗实用化快繁技术程序。

Abstract:Ribes nigrum varieties ----- Bopi and Houpi were adopted to carry out shoot ,tip culture in vitro. An average multiplication rate 4.7 shoots which were 1.4-3.1 times higher than that previously reported was achieved within 20 days by regulating BAP concentration in the medium. 83.3%-96% of rooting was obtained following 1-3 days of dark treatment with shoots at the density of 3 shoots per vessel on a half strength MS medium without growth hormones. With the substrate of vermiculite in wooden box, 97% of survival in. prickling plantlets was made. The plantlets were successfully - transplanted to the nursery with a vigorous grow th. A practical procedure for commercial plant production in Ribes nigrum by microproagation was established.
