Abstract:The study was to prepare a Agaricus blazei polysaccharide Lactobacillus acidophilus synbiotic that could pass through the acidic environment of gastric juice and be released in intestinal juice. Sodium alginate and Agaricus blazei polysaccharide were used as composite wall materials to wrap Lactobacillus acidophilus, and chitosan was used for secondary coating. The encapsulation rate, protection in artificial gastric juice and release in artificial intestinal juice were measured to evaluate the effect of microcapsules. Oxford cup was used to determine the bacteriostatic effect of synbiotics.The results showed that when the sodium alginate concentration was 1.0%, the Agaricus blazei polysaccharide concentration was 1.5%, the curing time was 4 min, and the prepared microcapsules had the best encapsulation rate,extremely higher than control group(P<0.01). After being exposed to gastric juice for 2 h, the viable count of synbiotic microcapsules was still 4.07×10;CFU/g. The maximum number of viable bacteria released by the three groups of microcapsules in the simulated intestinal fluid was at 2 h, and the differences among the groups were extreme(P<0.01).The antibacterial activity of ASL1 was better than that of Lactobacillus acidophilus solution(LA) and Agaricus blazei polysaccharide solution(AB). The experiment indicates that synbiotic microcapsules have better embedding effect and protective effect on Lactobacillus acidophilus, which can significantly improve the antibacterial activity.
