滇重楼根际土壤解无机磷细菌的分离与鉴定

摘    要:研究筛选滇重楼根际土壤解磷菌,提高滇重楼产量和品质,为其微生物菌肥的研发提供优良菌株。采集四川、云南、贵州中10个不同地区野生品和移栽品滇重楼根际土壤分离筛选高效解无机磷菌,通过钼锑钪比色法进行溶磷能力测定,结合生理生化和16S rDNA进行鉴定分析。结果表明,共分离纯化42株溶磷菌,发酵液中有效磷的含量为66.68~104.10 mg/L,其中属于芽孢杆菌属Z3-4菌株解无机磷能力最强,发酵上清液中有效磷的含量为104.10 mg/L、增磷量为38.55 mg/L、解磷率为37.03%。菌株Z3-4是一株具有高效解无机磷作用的细菌,可应用于生物菌肥的生产。

Abstract:The P-dissolving bacteria in rhizosphere soil of Paris polyphylla var. yunnanensis was screened in order to improve the yield and quality of Paris polyphylla var. yunnanensis,and provide excellent strains for the development of microbial fertilizer. The rhizosphere soils of wild and transplanted Paris polyphylla var. yunnanensis in 10 different regions of Sichuan,Yunnan and Guizhou were collected and screened to isolate and screen high-efficiency inorganic P-dissolving bacteria. The P-dissolving capacity was determined by the molybdenum-antimony-scandium colorimetric method,combined with physiological,biochemical and 16 S rDNA identification analysis. The results showed that a total of 42 strains of phosphate solubilizing bacteria were isolated and purified. The available phosphorus content in the fermentation broth was 66.68 ~ 104.10 mg/L. Among them,the Z3-4 strain belonging to the genus Bacillus had the strongest ability to solubilize inorganic phosphorus,and was excellent in fermentation. The content of available phosphorus in the clear liquid was 104.10 mg/L,the amount of phosphorus increased was 38.55 mg/L,and the phosphorus solution rate was 37.03%. It can be concluded that strain Z3-4 is a bacterium with high efficiency of dissolving inorganic phosphorus and can be used in the production of biological fertilizer.
