摘    要:本文比较了cDNA点杂交与酶联免疫吸附试验在大麦黄矮病毒的鉴定、检测及病害诊断中的应用特点及价值,并利用这两种技术研究在混合侵染中不同病毒(株系)间所发生的一些互作。通过使用适当的抗体或。cDNA探针,这两种检测技术均能特异地测定大麦黄矮病毒。尽管cDNA点杂交具有更高的灵敏度,但酶联免疫技术是目前最理想的病毒快速检测方法。这两种方法结合后所产生的另一技术-免疫杂交技术可成功地检测自然条件下受混合侵染病株内病毒或株系间的相互作用,这些相互作用包括"表型混合现象"和"反式壳体化"等。本文还对自然条件下检测这种病毒间相互作用的意义进行了讨论。

Abstract:ELISA, cDNA dot blot hybridization, and combination of the two techniques (i.e.immunohybridization) were used to detect different strains of barley yellow dwarf virus(BYDV) and their interactions in mixed infections. The results showed that both ELISAand cDNA dot blot hybridization could be used in detection of BYDV, though the latterwas more sensitive than the former technique. Because of its simplicity, however, ELISA (especially direct double-sandwich ELISA) was more suitable in disease diagnosis.Immunohybridization, which is a combination of some procedures of ELISA and cDNAdot blot, was very sensitive and useful in detecting interaction between BYDV strains in mixed infection, such as heterologous encapsidation (transcapsidation).
