单克隆抗体检测水稻白叶枯病菌的研究

摘    要:用ELISA双夹心法,以单克隆抗体为第二抗体,对60份水稻白叶枯病的病种和病叶进行检测,2张病叶在7ml0.01molPBS(pH7.2)中的浸出液稀释至32倍表现明显阳性;病种稻壳与PBS的比例(W/V)>1:3时效果明显。被测的病种稻壳浸出液的光密度值大于健种稻壳浸出液光密度值1.2倍可定为阳性。该法灵敏度为1.9μg/50μl。

Abstract:Application of ELISA method (Double sandwich method) in the tests of infected leaves and seeds by X. oryzae was carried out, it's McAb acted as the second antibody. The diseased leaves and seeds were immersed in PBS, and the immersion extracts used as the test sample containing bacteria of X. oryzae resulted positive, the method's sensitivity is related to the concentration of test samples, especially in the test of diseased seeds, our experiments indicated that the weight of seeds to the volume of PBS (W/V) should be in the ratio of one to three or more. By the analysis of significance of difference, as the O. D value of disease seeds immersion extract is more than 1.2 times than that of healthy seeds, the results could be regarded as nositive reaction.
