摘    要:本试验对携带不同目的基因和启动子的农杆菌菌株在去掉抗生素条件下进行菌液二次悬浮时的质粒稳定性进行了研究。结果表明:GV3101菌株中的几丁质酶基因质粒与AGL1菌株中的CBF3/rd29a质粒非常稳定,未发生质粒丢失现象;其次是LBA4404菌株中的CpTI质粒,虽发生个别质粒丢失,但与对照相比没有显著差异;携带CBF3/rd29a质粒的EHA105菌株悬浮培养12h后质粒发生明显丢失;含β-1,3葡聚糖酶基因质粒的EHA105菌株、含CBF3/CaMV35S质粒的GV3101菌株悬浮培养10h后质粒发生明显丢失;EHA105菌株CBF1/rd29a质粒和AGL1菌株CBF3/CaMV35S基因质粒最不稳定,分别悬浮培养6h和4h后即发生质粒明显丢失,且随悬浮培养时间的延长,质粒丢失率显著增高。质粒丢失程度与目的基因、启动子和农杆菌菌株三要素中的单一因素没有规律性联系,而是与目的基因、启动子和农杆菌菌株的组合有关;同时,在悬浮时间为0.5~6.0h范围内,质粒丢失主要与菌液浓度有关,而与悬浮时间关系不大;直接吸取一次悬浮液或离心后分别悬浮于细菌培养液和植物培养液等不同的悬浮培养方式下质粒稳定性没有差异。

Abstract:In this research genetic transformation plasmid stability of Agrobacterium tumefaciens was studied under the conditions of the second suspend liquid culture and without selective pressure of antibiotics,the tested plasmids harboured with different target genes and their promoters.The results showed that plasmids harboring Chitinase gene in GV3101 strain and plasmids harboring CBF3/rd29a gene in AGL1 strain both had much more stable,which no any plasmids lost during the second suspend liquid culture.Although...
