纤维素酶纯化方法与原生质体的制备

摘    要:以绿色木霉为材料,经稻草粉固体发酵、浸提得纤维素酶粗酶液,然后经硫酸铵分级沉淀,超滤,SephadexG-75柱层析3种方法纯化,利用得到的纤维素酶,分别制备幼嫩番茄叶片的原生质体。结果发现高度纯化的纤维素酶几乎得不到原生质体,而经超滤后的纤维素酶制备原生质体效果最佳。

Abstract:Trichoderma viride was used as the material in this experiment,cellulase was produced by solid state fermentation with the straw powder and crude cellulase was gotten by immertion,then by three different treatments -(NH4)2SO4 classified precipitate,ultrafilter and Sephadex G-75 chromatography. Three different treatments were done separately for the preparation of protoplasts of the young tomato leaves and the results were that the high purified cellulase got less protoplasts and cellulase by the ultrafilter had the best effect.
