花生肌动蛋白基因的克隆及序列分析

摘    要:以花生(Arachis hypogaeaL.)为材料,研究肌动蛋白(actin)基因序列。分离了高纯度RNA。以RNA为模板,以RT-PCR方法,克隆了花生actin基因cDNA序列,长度为384 bp,编码128个氨基酸残基。此cDNA序列(命名为Ahactin)与几种高等植物actin氨基酸序列相似性大于80%。

Abstract:The objective of this study was to examine the peanut(Arachis hypogaea L.) actin gene.High quality RNA were extracted from peanut and used as a template for cloning the actin gene cDNA by RT-PCR.Colonies with target foreign DNA inserts was identified by PCR.Recombinant pMD18-T plasmid was identified by restriction digestion and PCR,also sequence analysis.Peanut actin gene cDNA sequence,namely Ahactin,is 384 bp long,coding 128 amino acids.Sequence analysis shows that the deduced amino acid sequence of Ahactin shares more than 80% identity with the reported actin protein sequences.
