黄芪多糖除蛋白质方法与条件优化

摘    要:为研究Sevag法除黄芪粗多糖中蛋白质的最佳条件,并比较Sevag法和反复冻融法的除蛋白质效果,采用单因素试验方法,研究不同溶剂比例、用量、处理时间对蛋白质去除的影响。结果显示,Sevag法除蛋白质最佳条件为氯仿:正丁醇=4:1,多糖溶液:Sevag试剂=2:1,搅拌时间为25min。Sevag法与反复冻融法除蛋白质的最适次数为4次,前者除蛋白质的效果优于后者,从而优化了去除黄芪粗多糖中蛋白质的方法,可为黄芪多糖的研究与生产提供技术参考。

Abstract:In order to study the best conditions of the Sevag method for removing the proteins in Astragalus polysaccharide, and also to compare the Sevag method with repeated freeze-thaw method on protein removing, the single-factor experiment was conducted to study the effects of different ratios of solvent volume, dosages, and treating time on protein removing. The optimum conditions for protein removing with Sevag method were as follows, that is, chloroform:n-butyl alcohol = 4:1, sample ; Sevag reagent=2 : 1, and the stirring time was 25 min. The optimal repetition of the Sevag method and the repeated freeze-thaw method to remove the protein was four times, and the former was better than the latter. Thus, optimizing the method to remove the protein from Astragalus crude polysaccharide can provide some technical references for further researches and the production of Astragalus polysaccharide.
