摘    要:为了优选宫衣净酊的渗漉提取工艺条件,以主药红花有效成分羟基红花黄色素A(HSYA)含量和渗漉浸膏得率为考察指标,选择乙醇体积分数、溶媒用量、渗漉速度和浸渍时间为考察因素,采用L9(34)正交实验法对宫衣净酊渗漉提取工艺进行优化筛选.结果表明,宫衣净酊优选工艺为加入10倍量70%乙醇,浸渍24 h,以2 mL/(min·kg)进行渗漉提取.优选的工艺条件稳定、可行、有效成分浸出率高.

Optimization of the Percolation Extraction Process for Gong-Yi-Jing Tincture using Orthogonal Design Method

Abstract:To optimize the percolation extraction process of Gong-Yi-Jing tincture. The L9 (34) orthogonal trial was investigated to optimize the extraction process of Gong-Yi-Jing tincture. A general full factorial design with four factors (ethanol concentration, the volume of percolation liquid, soaking time and percolating speed) and each factor with three levels was implemented. HSYA (Hydroxysafflor yellow A) content and the extraction rate in the percolation extraction were taken as the measure index; HSYA is an effective component of principal drug safflower carthamus L. and was determined by high-performance liquid chromatography method. The result of the orthogonal trial showed that the optimized extraction condition was to use the soak the medicinal materials for 24 h with 10 times share of 70% ethanol and collect the percolation liquid by 2 mL/(min·kg). The optimized extraction process was stable, practicable and cost savings with a good yield of components in extract superior to the former extraction process.
