黑曲霉YF1菌株对烟嘧磺隆的降解条件研究

摘    要:黑曲霉(Aspergillus niger)YF1菌株是本实验室筛选得到的对烟嘧磺隆具有较强降解能力的菌株。为优化YF1菌株的降解条件,本试验分别对葡萄糖浓度、培养温度、初始pH和转速进行单因素试验,结果表明,YF1菌株降解烟嘧磺隆的最适条件为:葡萄糖浓度5g/L、培养温度30℃、初始pH值6.50和转速50r/min。本试验在研究其降解条件的同时,发现溶液的pH均出现下降,随后采用高效液相色谱检测草酸、L-苹果酸、α-酮戊二酸、乙酸、柠檬酸、D-苹果酸、琥珀酸的标准品,通过保留时间确定样品中的有机酸为草酸,但未检测出其它6种有机酸,因此,可以确定YF1菌株可以代谢葡萄糖产生草酸。本试验初步明确了黑曲霉YF1菌株对烟嘧磺隆的降解特性,为该菌株的应用奠定了基础。

Abstract:Aspergillus niger YF1,which was isolated in the laboratory,had the capability to degradate nicosulfuron.In order to optimize the degradation conditions of YF1,glucose concentration,culture temperature,initial pH and speed of incubator shaker were studied by the single factor test in this study,and the results showed that the optimum glucose concentration,culture temperature,initial pH,and speed of incubator shaker for nicosulfuron degradation by YF1 were respectively 5 g/L,30 ℃,6.50,and 50 r/min.Meanwhile,the decrease of pH was found in the medium Ⅲ.Then the standard samples of oxalic acid,L-malic acid,α-ketoglutaric acid,acetic acid,citric acid,D-malic acid and succinic acid were detected by high performance liquid chromatography,and oxalic acid was found in the medium Ⅲ via retention time,but other organic acids were not detected.This study preliminarily determined the degradation characteristics of nicosulfuron by YF1,which laid the foundation for the application of this strain.
