丙酮酸乙酯的微囊化与微囊中含量检测

摘    要:[目的]对丙酮酸乙酯进行微囊化研究,并建立微囊化丙酮酸乙酯的反相高效液相色谱检测方法。[方法]用喷雾干燥法对丙酮酸乙酯微囊化。检测方法为:色谱柱为Tigerkin ODS柱(4.6 mm×250 mm,5μm),流动相为丙酮和乙腈的混合液(V/V=3∶2),流速为1.0 ml/min,检测波长为210 nm。样品经过水溶解、超声、流动相破壁、过滤等处理之后进样检测。[结果]微囊粉末表面光滑,无塌陷无裂缝。丙酮酸乙酯浓度在0.3~1.8 mg/ml的范围内与峰面积积分值呈良好的线性关系(R2=0.999 2),回收率为98.05%,RSD=1.17%。[结论]丙酮酸乙酯微囊化方法可行,微囊化丙酮酸乙酯含量检测方法能准确快速的检测微囊中丙酮酸乙酯的含量。

The Microencapsulation of Ethyl Pyruvate and Content Determination of Microencapsuled Ethyl Pyruvate

Abstract:[Objective]Study on microencapsulation of ethyl pyruvate and establish a HPLC method for determination of Ethyl pyruvate in microencapsules.[Methods] Microencapsulation of ethyl pyruvate by spray drying.The assay was performed on a Tigerkin ODS column(4.6 mm×250 mm,5 μm) by UV derector at the wavelength of 220 nm with the mixture of acetone and acetonitrile(3∶2) as the mobile phase at the flow rate of 1.0 ml/min.The sample dissolved in aqueous solution.After ultrasound extraction,broken the wall of microencapsulated by the mobile phase,then it was filtered.Finally,it was injected to be determined by HPLC.[Results] Microencapsulated powder surface was smooth,no collapse,no cracks.The amount of ethyl pyruvate was linear with its area over the range from 0.3-1.8 mg/ml(R2=0.999 2),the average recovery was 98.05%,RSD Was 1.17%.[Conclusion]The mircoencapsulation method of ethylpyruvate was feasible,and the content detlermination method was simple and accurate for the ethyl pyruvate in microencapsulated.
