大花蕙兰组织培养的初步研究

摘    要:以茎尖、侧芽和茎段作为外植体,研究了大花蕙兰(Cymbidium grandi florum)的组织培养。结果表明,在MS培养基中,当细胞分裂素/生长素的比值为3.0时,愈伤组织的诱导率最高,可达60%。愈伤组织诱导及芽苗分化适宜的6-BA与IBA的浓度分别为1.5mg/L和0.5mg/L,而添加IBA0.1mg/L与6-BA0.5mg/L的1/2MS培养基对于外植体的继代增殖及壮苗生根较为有利。茎尖的总体诱导率高于侧芽和茎段,上部侧芽优于基部侧芽。

Abstract:Studies were carried out on tissue culture of XCymbidium grandiflorum,using terminal buds,lateral buds and stem sections as explants.The results showed that when the ratio of cytokinin to auxin is 3,the inducing rate of callus (60%)is the best.The optimal concentration of 6-BA and IBA is 1.5 mg/L and 0.5 mg/L respectively on induction of callus and differentiation of shoots,while 1/2MS medium with 0.1 mg/L IBA and 0.5 mg/L 6-BA is more beneficial to proliferation, strengthening plantlets and taking roots of explants.General inducing rate of terminal buds is higher than lateral buds and stem sections,and the inducing rate of the upper lateral buds was superior to that of the bottom ones.
