艾叶内生放线菌的分离、系统发育及相关特性研究

摘    要:从艾叶药用植物中共分离得到19株内生放线菌,对这些菌株分泌胞外淀粉酶、蛋白酶、脂肪酶和纤维素酶特性以及拮抗农作物病原菌和人类病原菌活性进行了研究,并对部分活性菌株进行了16SrDNA系统发育分析。结果表明:在这19株内生放线菌中,具有产胞外淀粉酶活性的菌株11株,产蛋白酶活性的菌株11株,产纤维素酶活性的菌株8株;拮抗活性试验显示,有2株菌对禾谷镰刀菌(Fusarium graminearum Schw.)、水稻胡麻斑病菌Bipolaris oryzae(Breda deHaan)Shoem]和肠球菌(Enterococcus sp.)具有拮抗活性,3株菌对耐青霉素类金黄色葡萄球菌(Staphyloccocus aureus)具有拮抗活性。对其中的3株菌进行了16SrDNA系统发育分析,该3株菌均属于糖霉菌属,菌株IF11、IXS和IF22均同时与Glycomyces mayteni YIM61331T、Glycomyces sambucusE71T、Glycomyces scopariae YIM56256T亲缘关系最近,序列相似性分别为98.3%、98.3%和98.1%。

Abstract:Nineteen strains of endophytic actinomycetes were isolated from Artemisiae argyi.The feature of excreting enzyme and the antagonistic activity against pathogenic fungi of crops and pathogenic bacteria of human about these strains was studied.16S rDNA phylogenetic analysis of some activated strains was carried out.The result showed that the number of strains which could produce amylase,protease and cellulase was 11,11,8,respectively.Antagonistic ability tests showed two strains have activity against Fusarium graminearum Schw,and Bipolaris oryzae(Breda deHaan) Shoem,Enterococcus sp.and three strains have activity against Staphyloccocus aureus of penicillin tolerance.Three activated strains were selected to carry out phylogenetic analysis,and the result based on 16S rDNA sequences about these three activated strains indicated that they belonged to Glycomyces.The strain IF11,IXS and IF22 were all similar to Glycomyces mayteni YIM61331T,Glycomyces sambucus E71T,Glycomyces scopariae YIM56256T,and the sequence similarities of 16S rDNA were 98.3%,98.3% and 98.1%,respectively.
