矮牵牛愈伤组织诱导及原生质体的分离与纯化

摘    要:研究了影响诱导矮牵牛(Petunia hybrid)形成愈伤组织和分离、纯化其原生质体效率的关键因素。结果表明,矮牵牛无菌苗叶片可在含1.0 mg/L 6-苄氨基腺嘌呤(6-BA)和0.1 mg/L萘乙酸(NAA)的MS培养基上形成愈伤组织,用酶液(2%纤维素酶R-10 + 0.5%果胶酶Y-23 + 0.3%离析酶R-10,0.5 mol/L甘露醇)对继代培养10~15 d的愈伤组织在黑暗条件下静置酶解12 h,原生质体产量和活性分别达到3.4×106个/g 和83%。

Abstract:Leaves of sterile Petunia hybrida seedlings were used to study the key factors affecting the efficiencies of callus induction and protoplast isolation.The results showed that the callus could be induced on the MS medium containing 1.0 mg/L 6-benzylaminopurine(6-BA) and 0.1 mg/L 1-naphthlcetic acid(NAA).Calluses subcultured in the same medium for 10-15 days were then hydrolyzed with solution containing 2% cellulase R-10,0.5% pectolyase Y-2,0.3% macerozyme R-10 and 0.5 mol/L mannitol in dark for 12 hours and the yield and viability of protoplasts reached up to 3.4×106/gram and 83%,respectively.
