摘    要:[目的]获得较透明的福尔瓦膜。[方法]采用两步法制备载网支持膜,再经电子束轰击形成超薄的福尔瓦膜,用于观察小鼠肝细胞的亚显微结构,并与常规福尔瓦膜进行图像效果比较。[结果]两步法制备的载网支持膜在电子显微镜下观察呈现多孔纤维状,经电子束反复照射,获得去除火棉胶的超薄福尔瓦膜。用两步法制备的超薄福尔瓦膜观察小鼠肝细胞的亚显微结构,图像清晰度高,比常规制备的福尔瓦膜图像效果好,差异较明显,具有统计学意义。[结论]两步法的膜制备方法可用于观察精度较高且要长期保存的样品。

Abstract:[Objective] The purpose was to get comparatively transparent Formvar membrane.[Method] The support film on carrying network was prepared by two-step method and then made into the ultrathin Formvar membrane by bombarding with electron beam,which was used to observe the ultrastructure of liver cell in mice and its image effect was compared with that of conventional Formvar membrane.[Result] The support membrane on carrying network prepared by two-step method and observed under electron microscope was fibrous-porous.The ultrathin Formvar membrane without collodion was obtained through repeated irradiation with electron beam.When the ultrastructure of liver cell in mice was observed with the ultrathin Formvar membrane prepared by two-step method,the image had high definition and its image effect was better than that of Formvar membrane prepared by conventional method,their difference was relatively obvious and had statistics meaning.[Conclusion] The two-step method of preparing membrane could be used in the observations of sample which needed higher precision and would be stored for long term.
