烟草中水溶性糖的分析方法研究

摘    要:[目的]研究烟草中水溶性糖测定的影响因素,建立烟草中水溶性糖的测定方法,为判断卷烟感官品质提供理论支撑。[方法]准确称取1.0000g样品,转移至具塞三角瓶中,加入纯净水,于40℃下超声萃取30min,过滤。利用液相色谱分析,考察了提取时间、提取温度、流动相、ELSD参数对水溶性糖测定值的影响。[结果]最终确定超声萃取,其中,提取时间30min,提取温度40℃,以乙腈-水为流动相。柱温30℃,蒸发光散射检测器漂移管温度80℃,载气流速2.00L/min。该方法中果糖、葡萄糖、蔗糖、麦芽糖在含量0.2~20.0μg范围内与其色谱峰面积之间具有良好的线性关系,其检出限分别为12、10、14、20ng,4种糖相对标准偏差均小于5%。[结论]该方法操作简单、方便,适用于烟草中水溶性糖含量的测定。

Abstract:[Objective] The research aimed to study the influencing factors of water soluble sugar determination in tobacco, set up the determination methods of water soluble sugar in tobacco and provide the theoretical support for judging the sensory quality of cigarette. [Method]1.000 0 g tobacco samples were transferred into a taper flask containing purity water, after ultrasonic extraction 30 min at 40 ℃ and filter, then the filtrate were analyzed by liquid chromatography with evaporative light scattering detector (HPLC-ELSD). The effects of extraction time, extraction temperature, mobile phase, ELSD parameter on the determined value of water soluble sugar were studied. [Result] The experimental results showed that ultrasonic extraction time was 30 min, temperature was 40 ℃, acetonitrile-water was used as the mobile phase and the column temperature was 30 ℃, the drift tube temperature of ELSD was 80 ℃ and the air flow rate was 2.00 L/min.The proposed HPLC-ELSD method had a good linearity from 0.2-20.0 μg of fructose,flucose, sucrose and maltose in the leaching solution, with the detection limits of 12, 10, 14,20 ng respectively. The relative standard deviation of the method for each kind of sugar was less than 5%.[Conclusion] The method is accurate, rapid and has been applied for the quantitative analysis of soluble carbohydrates in tobacco.
