蛹虫草超氧化物歧化酶的分离纯化及性质研究

摘    要:以蛹虫草为材料,经过硫酸铵盐析、SephedexG!75柱层析和DEAE!52柱层析,得到纯化的超氧化物歧化酶(SOD),经聚丙烯酰胺凝胶电泳显示单一蛋白区带,此酶比活力为17855.73U/mg,纯化倍数为53.7,回收率为21.8%。该酶在40℃保温150min,活性不变;在pH值5.0~9.0有较好的稳定性。该酶每亚基含1原子铁,对H2O2和乙醇-氯仿溶液敏感。

Abstract:Superoxide dismutase (SOD) from Chinese caterpillar fungus was separated and purified through ammonium sulfate precipitation andcolumn chromatographywith SephadexG-75 gel filtration and DEAE-52 ion exchange. The purified enzyme showed a single band on PAGE. Thespecific activity was 17 855.73 U/mg, the SOD enzyme was 57.3-fold purified, 21.8 % of the SOD activity was recovered. The SOD from Chinesecaterpillar fungus had good heat (40 ℃, 150 min) and acid- base stability in pH 5.0 ̄9.0, in which the activity of SOD did not decline. Everysubunit ofthis SODcontained one iron atom. The enzyme was sensitive toH2O2and Tsuchihashi.
