牛生长激素BGH基因的克隆及其真核表达载体pEGFP-N1-BGH的构建

摘    要:采用逆转录聚合酶链式反应(RT-PCR)技术,从11月龄的牛脑垂体组织中扩增出BGH基因片段,通过DNA重组技术将该基因片段重组于pEGFP-N1真核表达载体上,构建pEGFP-N1-BGH重组质粒。通过PCR扩增、酶切电泳分析和测序的方法对重组DNA进行鉴定,结果表明,成功构建了pEGFP-N1-BGH真核表达载体。

Abstract:The bovine growth hormone(BGH) gene was amplified from the tissue of calf brain pituitary by RT-PCR and cloned into the eukaryotic expression vector pEGFP-N1.The recombinants were further identified by PCR amplification,enzyme digestion and DNA sequencing.The results showed that the eukaryotic expression vector pEGFP-N1 was successfully constructed.
