艾草内生放线菌Ac10在植株体内定殖能力的测定

摘    要:为研究艾草内生放线菌Ac10的选择性培养基及在植株内的定殖能力和对植物生长的影响,测定了Ac10对4种抗生素(萘啶酸、利福平、氨苄西林、链霉素)和5种杀菌剂(啶酰菌胺、啶菌噁唑、苯醚甲环唑、杀毒矾、多抗霉素)的抗性,筛选出标记Ac10的药剂及其浓度,并以加入相应浓度药剂的培养基为基础,测定Ac10在玉米、番茄、白菜和黄瓜4种植株体内的定殖能力及对这些植物胚轴和胚根生长的影响.结果表明:Ac10能在含500 mg/L多抗霉素和25 mg/L萘啶酸或50 mg/L氨苄西林的培养基上正常生长;Ac10在同一植株的根茎叶中定殖能力显著不同(由强到弱依次为根、茎、叶,P0.05),在不同植株体内的定殖能力也显著不同(由高到低依次为玉米、番茄、白菜、黄瓜,P0.05);Ac10对玉米、番茄、黄瓜胚轴和胚根的生长无促进作用(P0.05),对白菜胚根的生长具促进作用(P0.05).可见,Ac10作为生防因子用于大田实践具有一定的可行性.

Abstract:In order to study selective medium, colonization ability in plants and the effect on plant growth of endophytic actinomycetes Ac10, four antibiotics (nalidixic acid, rifampicin, penicillin, and streptomycin) and five fungicides (boscalid, pyrisoxazole, difenoconazole, sandofan, and polyoxin) were used to determine the antibiotic or fungicide resistance of Ac10. And the colonization ability and the effect on growth of hypocotyl and radicle of Ac10 in corn, tomato, cabbage, and cucumber were tested based on the selective medium. The result showed that the Ac10 could suffer the 500 mg/L polyoxin and 25 mg/L nalidixic acid or 50 mg/L penicillin. The colonization ability of Ac10 in the root, stem, and leaf of one plant were significantly different (the root the stem the leaf,P0.05),and in different plant were also significantly different (the corn the tomato the cabbage the cucumber,P0.05). There was no variation in the growth of hypocotyl or radicle of tomato, cucumber and corn treated by Ac10 suspension (P0.05).But the growth of cabbage radicle treated by Ac10 suspension was significantly increased (P0.05).Thus,there is certain feasibility about Ac10 as a biocontrol agent applying in the field.
