人瘦蛋白基因小鼠遗传性的初步研究

摘    要:对转基因小鼠能否将外源基因遗传给后代以及如果能又将遵循怎栉产的遗传规律的问题进行了研究,在本研究中,2只转基因母鼠与非转基因公鼠交配,妊娠足月分娩后分别得到7只和8只仔鼠,以DIG标记的人瘦蛋白基因为探针,进行Southern杂交,结果表明在左只转基因母鼠生出的仔鼠中各有3只为含有转基因片段的仔鼠,这表明转基因可以遗传给后代,统计分析结果初步表明,外源基因的遗传遵循盂德尔遗传方式。

Abstract:In this article,the following two questions was answered: (1) Could transgene(s) be transmitted to the offspring of the transgenic animals? And (2) if could, how to inherit? Two female transgenic mice were mated with nontransgenic male mice. After full term pregnancy and parturition, 7 and 8 baby mice were produced by the two female transgenic mice respectively. Every 3 among the 7 and 8 mice were confirmed to have the transgene(s) in their genome by Southern hybridization with DIG labelled human leptin gene (cDNA ) as probe. It showed that transgene(s) could be transmitted to their offspring of the transgenic animal.The statistical results showed that the transgenes inheritance followed the Mendelian pattern.
