鰤诺卡氏菌灭活疫苗对大口黑鲈非特异性免疫指标的影响

摘    要:研究大口黑鲈(Micropterus salmoides)对鰤诺卡氏菌(Nocardia seriolae)灭活疫苗非特异性免疫应答的变化过程,为预防鰤诺卡氏菌病提供理论依据,可促进养殖业健康发展。大口黑鲈平均体长(10±2)cm、平均体重(30±2)g,鰤诺卡氏菌从患病大口黑鲈身上分离鉴定获得。600尾大口黑鲈随机分为免疫组和对照组,每组300尾,免疫组通过腹腔注射菌浓度为1.0×109CFU/mL的鰤诺卡氏菌灭活疫苗0.5 mL/尾,对照组注射同等剂量的磷酸盐缓冲液。免疫后的第1、7、14、21、28和35天采集血液样品,测定血细胞数量、吞噬细胞吞噬活性、溶菌酶活力、酸性磷酸酶活力以及超氧化物歧化酶活力等非特异性指标。免疫接种第35天后进行攻毒试验,计算相对免疫保护率。结果表明:免疫组血细胞数量显著增加,红细胞和白细胞数量均于第7天达峰值,分别为(4.49±0.23)×106个/mm3和(3.88±0.24)×103个/mm3;免疫组吞噬指数和吞噬百分比均在第7天达到最高值...

Abstract:InSthisSstudy,the SNocardiaSseriolae whole cell vaccine inactivated by formalin was used to immunize healthySMicropterus salmoides by peritoneal injection.The blood samplesSofSMicropterus salmoideSatS1,7,14,21,28 and 35 days post immunization were collected and the non-specificSindexesSsuchSasSblood cell count,phagocytic activity of phagocyticScells,lysozymeSactivity,acidSphosphataseSactivity,superoxide dismutase activitySwereSmeasured.InStheSimmune group,theSnumberSofSbloodScellsSincreasedSsignificantly,and the numbers of erythrocytes and leucocytes both reached the maximum values of (4.49±0.23)x106/mm3 and (3.88±0.24)x103/mm3,respectively,at 7d post immunization.The phagocytic index and phagocytic percentage reached the maximum valuesSatS7d post immunization,which were 2.65 and 29.85%,respectively.The activity of lysozyme and acid phosphatase reached the maximum values at 7d post immunization,while the superoxide dismutase activity reached its maximum value at 14d post immunization,which were all significantly higher than those of the control group (P<0.01).The challenge infection test showed that the relative immunity protection rate was 61.54%.These results showed that the inactivated Nocardia seriolae vaccine could significantly induce the immune response of Micropterus salmoide improve the activity of non-specific immune factors,enhance the immune protection of fish.
