大豆幼嫩子叶离体培养及辐照处理配合基因枪遗传转化研究

摘    要:3年来共收集 40份栽培大豆和野生大豆材料 ,进行幼嫩子叶的离体培养 ,其中 3个品种获得较高的再生植株频率。辐照处理配合基因枪介导转基因研究表明 ,5Gyγ射线辐照处理能明显提高抗性愈伤的频率。GUS检测抗性愈伤组织 ,证明外源基因已导入大豆细胞 ,并在其中表达

IN VITRO CULTURE OF SOYBEAN IMMATURE COTYLEDONS AND GENETIC TRANSFORMATION VIA BIOLISTIC SYSTEM
Abstract:soybean ( Glycine max (L.)Merr.) cultivars were collected to study in vitro culture of immature cotyledons.Regenerated plants of 3 cultivars were obtained and one cultivar was used to study the effect of irradiation treatment on stable genetic transformation rate.0,5,10,20 Gy were applied before or after bombardment with Bio Rad system.It was found that 5 Gy was the effective dose to raise the ratio of cotyledons with Km R calli.GUS enzyme activity showed that foreign genes had been integrated into genome of soybean callus.
