摘    要:目的以优化金银忍冬花粉离体萌发的培养基组分。方法以盛花期新鲜花粉为材料,采用花粉离体萌发方法,在单因素试验的基础上,通过响应面法优化培养基组分。结果金银忍冬花粉离体萌发的最佳培养基组分为蔗糖质量浓度为400g/L、硼酸质量浓度为127.23mg/L、2,4-D质量浓度为75.00mg/L,在此培养基组分条件下,金银忍冬花粉萌发率的预测值为90.24%,验证萌发率为89.65%。结论利用响应面分析法可得到理论最佳培养基组分,验证结果表明该方法科学、高效。

Optimization on Medium Components of Pollen Germination in Vitro in Loniccera maackii(Rupr) Maxim

Abstract:Objective The purpose of this experiment was to optimization of medium components of Loniccera maaekii(Rupr) Maxim. Method The experiment used fresh pollen as material, through the method of pollen germination in vitro, Based on single-factor Experiments, response surface methodology was used to optimize the medium components. Results The optimum medium compositions of pollen germination were found as follows: sucrose concentration was 400g/L, boric acid concentration was 127.23mg/L,2,4-Dconcentration was 75.00mg/L, Under these conditions, the predicted and experimental pollen germination yields were 90.24% and 89.65%. Conclusion Response surface method can provide a approach to find optimal conditions for pollen germination. The results suggest that the new method is effective and economical.
