蝴蝶兰愈伤组织诱导和体细胞胚胎发生的研究

摘    要:研究了蝴蝶兰愈伤组织的诱导和通过体细胞胚胎发生的植株再生。原球茎在含有蔗糖的VW培养基上形成愈伤组织。结果表明,最有效的诱导愈伤组织的培养基为VW+0.1 mg/L BA+0.1 mg/L 2,4-D+60 g/L蔗糖或者VW+0.3 mg/L BA+0.1 mg/L NAA+40 g/L蔗糖。愈伤组织在被转到不含植物激素的VW培养基上易形成原球茎。组织学观察证实了体细胞胚胎结构的形成,暗示了试验中诱导的愈伤组织即为体细胞胚。

Abstract:Callus induction and plant regeneration through somatic embryogenesis in Phalaenopsis were researched.Protocorm-like body(PLB)segments formed callus on VW medium with sucrose.The results showed that the medium containing 0.1 mg/L BA,0.1 mg/L 2,4-D and 60 g/L sucrose or 0.3 mg/L BA,0.1 mg/L NAA and 40 g/L sucrose was effective for callus induction.The callus easily formed PLBs after being transferred to VW medium without plant hormone.Histological observation proved that the somatic embryo structure formation and suggested that the callus induced in this work was somatic embryos.
