茶树紫色芽叶中花青素的提取-层析分离及初步鉴定

摘    要:[目的]为茶叶中花青素的提取、分离与鉴定提供方法支持。[方法]以茶树紫色芽叶为原料,通过提取、除杂获得花青素粗提物,再通过柱层析和薄层层析分离获得一种花青素,采用紫外-可见光谱法对其结构进行鉴定。[结果]花青素粗提物在465~550 nm波长范围内有花青素特有的吸收峰,可以初步确定花青素粗提物中含有花青素。花青素粗提物在浓盐酸中显粉红色,且其在KI·I_2试剂的作用下不产生棕色沉淀,表明花青素主要存在于花青素粗提物中,而且其中不含有咖啡碱。显色鉴定结果表明,柱层析出的F8含有花青素。显色和紫外-可见光谱法的鉴定结果表明,薄层层析出的C2为花青素组分。以紫芽茶为原料,采用乙醇水溶液提取花青素,并通过石油醚脱脂,三氯甲烷、乙酸乙酯除杂获得花青素粗提物,所得花青素产率为6.44%。[结论]该研究对茶树资源的开发利用具有重要意义。
