星油藤根腐病的症状和病原鉴定

摘要:采用切片和组织分离法对星油藤根腐病病原菌进行分离,结合形态学观察与分子生物学方法对病原菌进行鉴定,按柯赫氏法则对其致病性进行测定。结果表明:PCR技术扩增病菌rDNA-ITS基因,获得长度为550~650 bp的DNA片段;菌株序列与尖孢镰刀菌的序列同源性达到99%;星油藤根腐病的病原菌为尖孢镰刀菌(Fusarium oxysporum),通过重分离获得该菌株。

Abstract:The pathogen of Plukenetia volubilis root rot was isolated using section-cutting and tissue isolation method, identified by morphological observation and molecular biology method, and determined according to the Koch′s rule. The result showed that rDNA-ITS gene was amplified by PCR technique, DNA fragment with a length of 550-650 bp was obtained. The sequence homology between the stain and Fusariun oxysporum was 99%. The pathogen of P.volubilis root rot was F.oxysporum.
