转精氨酸酶基因ZmARG玉米株系AKF65的杂交组合鉴定及遗传稳定性分析

摘    要:以转玉米精氨酸酶基因ZmARG的T_4代株系AKF65为试材,配置杂交组合,对组合进行农艺性状调查和产量性状鉴定,并将F_1进行回交,采用PCR分子检测手段分析ZmARG基因的遗传特性。结果表明,杂交组合合344×AKF65和WY-1-2×AKF65在株高、穗位高等性状上,与非转基因对照组合相比差异均不显著,在产量、单穗重、穗长等性状上显著高于对照组合,百粒重、粒宽和粒厚等性状上显著高于常规对照品种克玉15。BC1F1回交世代中ZmARG基因分离比例均接近1∶1,表明外源ZmARG基因为显性并稳定插入到基因组中。

Abstract:High yield transgenic maize lines AKF65 with ZmARG at T4 generations were used to cross with He344 and WY-1-2. The phenotypic traits and yield traits of hybrid combinations were identified, and the genetic stability was analyzed by PCR test using BC1F1 population. The results showed that there was no significant difference in plant height and ear height between the two transgenic hybrid combinations and control. The plot yield, single panicle weight and ear length of He344×AKF65 and WY-1-2×AKF65 were significantly higher than control. The hundred-grain weight, grain width and grain thickness of the two transgenic hybrid combinations He344×AKF65 were significantly higher than Keyu15. The separation ratio of ZmARG gene in BC1F1 population was close to 1:1. It indicated that the ZmARG gene was dominant and stable integrated into AKF65 genome.
