沙棘离体快速繁育技术研究

摘    要:本项研究基于组培手段,试图解决利用沙棘茎段快速培养其完全苗的问题。通过对沙棘外殖体消毒、褐变的防止、芽的增殖、生根诱导等过程的系列试验表明:在沙棘外殖体的供试消毒液中,消毒效果以0.1%升汞与20%酒精混合液为佳;用200ppmVc浸泡材料5h是抑制沙棘外殖体褐化的有效方法;芽增殖过程效果较好的是1/2B5 IAA0.3-0.5培养基。该结果为沙棘离体快繁提供了一定的经验和技术支撑。

Abstract:This research resolved the problem in the rapid propagation of Seabuckthorn stems by tissue culture. Experiments of explant sterilization, browning prevention, sprout proliferation, root inducement on tissue culture of Seabuckthorn were studied. The results showed that the 0.1% HgCl and 20% ethanol solution gained the best sterilize effect. Marinating 5 hours in Vc solution(200ppm) was effective to prevent explant browning. The culture medium including 1/2B 5+IAA 0.3-0.5 was effective for sprout proliferation. These results would be useful for the rapid propagation of Seabuckthorn.
