摘 要: | 以生产上大面积推广的小麦品种豫麦18-64、豫麦34号和豫麦70号为供试材料,用基因枪介导法进行了抗除草剂基因(Bar)的转化,研究了幼胚不同的预培养时间和渗透处理对转化效果的影响。结果表明,幼胚以诱导培养10~15d的转化效果最好,在此基础上结合用0.4mol/L的甘露醇在轰击前6h和轰击后18h进行甘露醇处理,可明显提高转化效率。

Effect of the Growth Appearance of Acceptors on the Frequency of Wheat Transformation by Microprojectile Bombardment |

Abstract: | In this paper, the acceptors were from 3 wheat cultivars including Yumai 18-64, Yumai 34 and Yumai 70. The plasmid with resistant herbicide gene(Bar) was transferred by microprojectile bombardment. The factors affecting the Biolistic transformation like acceptor' s pre cultural time and its treatment were studies. The result showed that the immature embryos pre cultured for 10~15 days and treated with 0.4mol/L mannitol could significantly improved gene transformation frequency. |
