摘    要:根据GenBank中猪流行性腹泻(PED)疫苗株与野毒株ORF3的特点设计特异性引物,建立能够快速区分PEDV疫苗株与野毒株的RT-PCR检测方法。建立的RT-PCR检测方法能够对PEDV疫苗株与野毒株扩增出特异性片段,其大小分别为278 bp和327 bp。该方法具有快速、特异、通用等特点,可用于实验室快速诊断、野毒株的区分,为该病的诊断及免疫防控提供参考依据。

Establishment and application of RT-PCR assay for detection of PEDV vaccine and wild strains
Abstract:Based on the characteristics of porcine epidemic diarrhea (PED) gene in ORF3 between vaccine and wild strains, one pair of primers was designed and a rapid diagnostic method of RT-PCR was established. The established method had a rapid and general features and it only amplified target band from PEDV. A total of 278 bp bands were amplified from vaccine strains and 327 bp bands were amplified from wild strains. A highly special RT-PCR method was established and it can directly differentiate PEDV vaccine strains and wild strains, which can be provide reference for diagnosis and the immune prevention.
