丹参对溃疡性结肠炎患者外周血中性粒细胞的凋亡、O-2和NO产生的调节作用

摘    要:目的:观察丹参(SM)对溃疡性结肠炎患者外周血中性粒细胞(PM N)凋亡及超氧阴离子(O2-)和一氧化氮(NO)产生的影响。方法:选择中度活动期溃疡性结肠炎(UC)患者15例(UC组)和健康体检者18例(对照组),采用人外周血PM N体外培养细胞模型进行实验研究,采用流式细胞术检测PM N凋亡,以细胞色素C还原法、化学比色法分别检测PM N产生O2-和NO的量。结果:UC组外周血PM N凋亡率明显低于对照组(P<0.01),产生O2-和NO的量高于对照组(P<0.01)。在体外丹参明显促进UC患者PM N凋亡(P<0.01),在500 m g/L内呈剂量依赖性;同时丹参可抑制UC患者PM N释放O2-、合成NO。结论:SM对体外PM N的凋亡及其产生O2-和NO有调节作用。

Abstract:Objective: To observe the effect of Salvia miltiorrhiza(SM) on the apoptosis and the production of superoxide anion(O~-_2) and nitric oxide(NO) in peripheral blood neutrophils(PMN) from patients with ulcerative colitis(UC).Methods: The PMN taken from UC patients at the middle active stage were cultured in vitro,then the apoptosis of PMN was determined with flow cytometry and the production of O~-_2 and NO in PMN were tested with cytochrome C reduction method and colorimetry,respectively.Results: The percentage of apoptosis of PMN in UC was obviously lower than that of control but the production of O~-_2 and NO were higher than those of control.SM injection solution promoted the apoptosis of PMN and inhibited the release of O~-_2 and the synthesis of NO in PMN.Conclusion: SM has the regulatory effects on the apoptosis and the production of O~-_2 and NO in PMN from the patients with UC.
