抗应激中药冲剂对猪肝脏热应激蛋白70表达的影响

摘    要:研究抗应激中药冲剂对高温应激下猪肝脏组织热应激蛋白70(Hsp70)表达的调控效果。选取64头3月龄中国实验用小型猪,随机分为常温对照组、高温应激组、方Ⅰ、Ⅱ组,于高温第1,3,6,10天动态取样后,分别采用免疫组织化学和ELISA法测定肝中Hsp70的蛋白表达定位定量变化。结果表明,高温应激后,猪肝脏组织细胞阳性率显著增加(P<0.05),方Ⅰ、方Ⅱ组肝脏细胞阳性率显著高于常温组(P<0.05)但低于高温组(P<0.05);ELISA方法结果显示高温组、方Ⅰ、方Ⅱ组肝脏组织Hsp70含量显著增加(P<0.05),方Ⅱ组Hsp70含量在第3,6天比高温组显著增加(P<0.05)。因此,中药抗应激冲剂可通过增加肝脏Hsp70蛋白含量水平来提高机体细胞热耐受,达到保护作用,且方Ⅱ组效果优于方Ⅰ组。

Abstract:The Anti-stress pulvis of Chinese herb on expression of Hsp70 in pig liver after heat stress.was invesfigated Sixty-four Chinese experimental miniature pigs aged 3 months were divided into 4 treatments in random,i.e.control,heat stress,PulvisⅠand PulvisⅡ groups.The livers were collected at the 1st,3rd,6th,10th day after heat stress and the expressions of Hsp70 in liver were detected by immunohistochemistry and ELISA.The results showed that,The Hsp70 average positive rates in cytoblast increased(P<0.05) after heat stress.The positive rates in PulvisⅠand PulvisⅡ were higher remarkably(P<0.05) than that of control group but lower(P<0.05) than heat stress group.The result of ELISA showed that the expressions of Hsp70 obviously increased(P<0.05) in heat stress,PulvisⅠand PulvisⅡ groups.The expression of PulvisⅡ was higher(P<0.05) than those in heat stress at 3rd and 6th day.The data showed that the Anti-stress Pulvis from Chinese herb could elevate the tolerance of cell to heat stress and protect cell through the increase the expressions of Hsp70,and the effect in PulvisⅡ was superior PulvisⅠ.
