白术茎尖的离体快繁研究

摘    要:目的]建立白术茎尖的离体快繁体系,筛选最佳培养基。方法]以白术的茎尖为外植体,设置7种附加NAA、IBA、6-BA不同浓度配比的MS培养基进行茎尖诱导分化与增殖培养,设置5种附加不同浓度NAA的1/2MS培养基进行生根培养,将驯化后的试管苗置于草木灰与草炭土(1:2)的混合基质进行移栽试验。结果]7种诱导分化与增殖培养基中,MS+6-BA0.4mg/L+NAA0.1mg/L的培养基有利于白术芽的萌发,芽成活率达100%;MS+6.BA0.5mg/L+NAA0.1mg/L的培养基有利于诱导丛生芽增殖。5种生根培养基中,以1/2MS+NAA0.1mg/L+活性炭0.5g/L诱导白术生根的效果最佳,生根率达66.7%。驯化后的试管苗在草木灰和草炭土的混合基质中培养,试管苗成活率高。结论]该研究为白术实现工厂化育苗及药用次生代谢产物研究提供了试验依据。
