摘    要:电镜研究发现,简易机械法分离得到的腔前卵泡有1~3层颗粒细胞,卵泡外层为颗粒细胞,包有一层厚的糖蛋白基底膜,外无卵泡膜,其颗粒细胞及卵母细胞超微结构特征与卵巢皮质片中的腔前卵泡一致。卵母细胞为球形或卵圆形,有一大而偏离中心的核,核内有一显著核仁,各细胞器均匀分布于细胞质中,卵母细胞中有大量线粒体及其它细胞器遍布卵胞质,卵母细胞质膜上有突起将卵母细胞表面的微绒毛与邻近的颗粒细胞相连。超微结构研究结果表明,本法获得的牛腔前卵泡没有受到分离过程的机械伤害。

Abstract:The transmission electron microscope(TEM) analysis showed that the mechanically isolated preantral follicles were with 1 to 3 layers of granulose cells tightly enclosing the oocyte and they were surrounded by a thick layer of glycoproteins,the basement membrane.There was no follicular interna theca around them.The ultrastructural characteristics of the preantral follicles isolated freshly were mostly similar to those presented in situ.The quiescent oocyte was spherical or oval,having a large eccentrically located nucleus with a conspicuous nucleolus.The organelles were uniformly distributed in the cytoplasm.The cytoplasm of oocytes also contains numerous rounded mitochondria and Endoplasmic Reticulum(SER,RER) besides the usual organelles.The oocyte plasma membrane presented projections that penetrated between adjacent granulosa cells and a few short microvilli lying parallel to the oocyte surface.The results indicated that this isolation procedure induced no damage to follicles.
