豆类丝核菌次级代谢产物与阿霉素的体外联合抑瘤作用研究

摘    要:[目的]研究豆类丝核菌次级代谢产物与阿霉素的降毒增效作用。方法]分别采用6种不同浓度的豆类丝核菌次级代谢产物(苦马豆素含量分别为10.00、5.00、2.50、1.25、0.62、0.31 μg/ml)处理人肝癌细胞HepG2,测量其IC50;然后用此代谢产物与2种不同浓度(60、6 μg/ml)阿霉素(ADR)联合处理HepG2,分析两者的增效作用。结果]豆类丝核菌次级代谢产物可抑制肝癌细胞HepG2生长,其对HepG2细胞作用72 h的IC50为1.25 μg/ml;ADR可显著抑制肝癌细胞HepG2生长,加入豆类丝核菌次级代谢产物后HepG2细胞的形态和存活率变化不大。结论]豆类丝核菌次级代谢产物与阿霉素体外联合不具有增效作用。

Abstract:[Objective] The aim was to study the synergism of secondary metabolites of Rhizotonia legumeinicola and adriamycin. [Method] The secondary metabolites of R. legumeinicola with different concn. (10.00, 5.00, 2.50, 1.25, 0.62, 0.31μg/ml) was taken to treat HepG2 and its IC_(50) was determined. And then the HepG2 cells were treated by the secondary metabolites combined with ADR with different concn. (60, 6 μg/ml), and their synergism were studied. [Result] The secondary metabolites of R. legumeinicola could inhibit the growth of HepG2 and its IC_(50) on HepG2 after 72 h was 1.25 μg/ml. The ADR could significantly inhibit the growth of HepG2, and the morphology and survival rate of HepG2 cells changed little after addition of the secondary metabolites of R. legumeinicola. [Conclusion] The secondary metabolites of R. legumeinicola combined with adriamycin had no synergism in vitro.
