金黄色葡萄菌球菌毒力基因在传代过程中的稳定性

摘    要:为了解分离自奶牛乳房炎的金黄黄葡萄球菌(Staphylococcusaureus)毒力基因在传代过程中的稳定性,对传代前后金黄色葡萄球菌的毒力和毒力基因进行测定和检测。本研究采用急性毒性试验的方法测定8株金黄色葡萄球菌原代、第5代、第10代、第15代、第20代及第25代的菌株对小鼠的半数致死量,通过聚合酶链式反应(PCR)检测原代、第5代、第10代、15代、第20代及第25代的菌株中5种毒力基因ZFB、ALF、BET、xjngnl及Nuc的变化情况。试验结果表明,金黄色葡萄球菌在传代过程中毒力基因分布较稳定;但Xingm、ALF、BET基因极易发生变异。经小鼠腹腔连续多次传代后毒力逐渐增大,可推知Xjngm毒力基因与毒力相关性最大,并易受传代影响。

Stability of Virulence Genes in Staphylococcus aureus Strains During Subculture

Abstract:To study the stability of virulence genes of S aureus strains which was isolated from mastitis during subculture, toxicity and toxicity gene were detected before and after subcul- tured. 8 strains original, the 5th generation, the lOth generation, the 15th generation, the 20th generation and the 25th generation S. aureus were administered to mice in order to detecte the me- dian lethal rate. Variation of Vir genes including ZFB, ALF, BET, Xjngm and Nuc were investigated by PCR. The results showed that the vir gene distribution kept stable during passage. But Xjngm, ALF,BET genes mutated extremely. Toxicity increased after passed many times from mice, which indi- cated that Xjngm has the most dependability to toxicity, and is easily be affected by subculture.
