蔓马缨丹挥发油GC-MS分析及其生物活性

摘    要:蔓马缨丹是我国华南地区常见的园林绿化灌木。为分析和鉴定蔓马缨丹挥发油的化学组成及其抗菌和杀线虫活性,本文采用水蒸汽蒸馏法提取蔓马缨丹枝叶的挥发油,通过GC-MS分析其化学组成,并分别测定其抗细菌和抗线虫活性。结果表明:蔓马缨丹枝叶挥发油的含量0.075%(以鲜重为基础),通过气相色谱与质谱联用技术共分析并鉴定出39种化学成分。进而采用峰面积归一化法推算得到各化合物的相对含量,其中含量最高的成分为β-Cubebene(19.94%)和β-Elemene(19.61%)。抗细菌活性结果表明,蔓马缨丹挥发油对桉树青枯病菌、番茄疮痂病菌以及黄瓜角斑病菌均表现出一定程度的抑制活性,其中对番茄疮痂病菌的生长抑制效果相对最好,其抑菌圈直径为30.40 ± 2.78 mm。另外,蔓马缨丹挥发油对南方根结线虫表现出了一定的致死活性,在24小时内的致死中浓度为33.07 ± 2.12 mg/mL。本研究结果为蔓马缨丹资源的综合开发与利用提供思路,为植物源农药的开发奠定了理论依据。

Abstract:Lantana montevidensis is a common landscaping shrub in South China. This study was to analyze and identify the chemical composition and antibacterial and nematicidal activities of the volatile oil of Lantana montevidensis. The volatile oil was extracted by hydro-distillation. Its chemical composition was analyzed by GC-MS, and its anti-bacterial and anti-nematode activities against four bacteria and two nematodes were determined respectively. The results show that the yield of volatile oil from L. montevidensis were 0.075% (w/fw), and thirty-nine components were identified from the volatile oil. The relative percentages of different chemical components was measured by peak area normalization, and β-cubebene (19.94%) was the highest component followed by the β-elemene (19.61%). The results of antibacterial activity showed that the volatile oil of L. montevidensis exhibited certain inhibitory activity against Ralstonia solanacearum, Xanthomonas vesicatoria and Pseudomonas lachrymans, and the diameter of inhibitory zone against X. vesicatoria reached 30.40 ± 2.78 mm. In addition, the volatile oil exhibited definite nematicidal activity against Meloidogyne incongnita, with the LC50 of 33.07 ± 2.12 mg/mL within 24 hours. These results would provided ideas for the comprehensive development and utilization of L. montevidensis resources and laid a theoretical basis for the development of plant-derived pesticides.
