勒杜鹃无菌系的建立与试管开花诱导的研究

摘    要:用勒杜鹃茎段为外植体,研究不同的外植体的接种方式与不同激素浓度配比对无菌系建立的影响,在此基础上进一步研究诱导勒杜鹃无根苗试管开花的外源激素浓度组合、蔗糖与琼脂浓度的组合,以及光照、温度等培养条件.结果表明:将茎段纵切,除去1/3组织水平接于MS+6-BA 0.05 mg/L培养基中,增殖系数最高,达到4.06;自然光照下,pH值=5.6,蔗糖浓度为3.0%,琼脂浓度为0.6%,无根苗在MS+6-BA 0.05 mg/L培养基中诱导试管开花时,开花诱导率最高,达到45.83%.
Abstract:
Sterilized sections of adventitious shoots induced from the axillary buds of Bougainvillea spectabilis Willd. were used as explants, and the effects of various inoculation modes and hormone combinations on the establishment of a germfree system were studied. Based on the results, the optimal culture conditions for in vitro flowering were investigated. The highest multiplication rate (4.06) was achieved when the explants were vertically sectioned and inoculated on MS+6-BA 0.05 mg/L. The highest in vitro flowering induction (45.83%) was obtained when rootless plantlets were cultured on MS + 6-BA 0. 05 mg/L,under natural lighting and temperature with a pH of about 5.6, a sucrose concentration of 3.0% and an agar concentration of 0.6 %.
