云南39个野生蔷薇种间遗传多样性的SSR分析

摘    要:利用简单重复序列SSR(Simple Sequence Repeat)标记技术对蔷薇属39份野生材料的遗传多样性进行了研究.用筛选出的20对SSR引物对39份材料DNA进行PCR扩增,在20个位点上共检测到249个等位基因,每一位点的等位基因变幅为6~19个,平均12.5个.材料间遗传相似系数变化范围为0.142~0.800,表明39个云南野生蔷薇种间具有丰富的遗传多样性.本研究发现,在相似系数为0.34时,基于SSR标记进行UPGMA的聚类分析将39份蔷薇野生种分为10个组,与植物形态学分类结果大体一致.
Abstract:
The genetic diversity of 39 Rosa wild species was studied by SSR (Simple Sequence Repeat).Twenty pairs of primers were used in PCR amplification with genomic DNA as template. A total of 249 alleles were detected at 20 loci. The number of alleles per locus ranged from 6 to 19, with an average of 12.5. The genetic similar coefficient ranged from 0. 142 to 0. 800, which showed that great genetic diversity existed among the 39 Rosa wild species in Yunnan based on SSR molecular markers. UPGMA cluster analysis distinctly classified the 39 wild Rosa species into ten groups at the similar coefficient 0.34, which is consistent with botanical morphological classification.
