Abstract:Elevated expression of Rubisco protein is one of the characteristics of plants responding to abiotic stress.In the previous proteomic analysis,the homologous clone was used to clone the differentially expressed gene GhRBCO of upland cotton’KK1543’under drought stress.The coding region of GhRBCO gene is 747 bp in length and encodes 249 amino acids.The alignment of amino acid sequences and homology analysis indicates that GhRBCO is highly consistent with RBCO proteins in other plants,indicating that the gene is quite conserved during evolution.The phylogenetic tree results showed that GhRBCO had the highest homology with Gossypium raimondii,Asian cotton and Artemisia annua.The expression level in the roots,stems and leaves of cotton showed a trend of slowly increasing and then decreasing,and the expression level in the roots reached the highest for the first time at 24 h.The lowest expression was found in stem at 12 h.The expression level of this gene was up-regulated in cotton roots,stem and leaves.Experiments show that GhRBCO gene may be involved in cotton regulation of abiotic stress,which lays a foundation for further study of GhRBCO gene function.
