Abstract:Brassica napus is an important oil crop in China and even the world.It is affected by diseases and insects,resulting in a decline in output.NBS-LRR genes have been proven to be broad-spectrum disease resistant genes,which are involved in the disease resistance to a variety of pathogens in Arabidopsis thaliana,rice.Two NBS-LRR differentially expressed genes,named BnRRS1-like and BnRRS2-like,were identified by transcriptome analysis of high/low oil near isogenic lines.The phylogenetic tree and blast analysis showed that BnRRS1-like and BnRRS2-like genes were shared homologous with Arabidopsis RRS1 genes.NCBI was used for comparative analysis of homologous sequences,and it was found that BnRRS1-like gene and BnRRS2-like gene were 60.68%and 52.39%similar to Arabidopsis RRS1-R gene respectively.The analysis of the conserved domains showed that both the BnRRS1-like(BnaC09 g18980)and BnRRS2-like protein(BnaA01 g01460)contained the conserved domain ofTIRNBSLRR,which belonged to the resistant protein of TIR-NBS-LRR.Conservative motif analysis revealed that BnRRS1-like and BnRRS2-like had very similar conserved motifs.Chromosomal localization showed that BnRRS1-like and BnRRS2-like genes were located at chromosome C9 and A1 respectively.Gene expression in root,stem,leaf and flower of Brassica napus was analyzed by qRT-PCR.The results showed that the expression level of the two genes was highest in the leaves of rape,followed by the roots,and lowest in the flowers.After Sclerotinia sclerotiorum inoculation,the relative expression levels of BnRRS1-like and BnRRS2-like genes were measured at different induction times.It was found that the expression levels of BnRRS1-like and BnRRS2-like genes increased significantly three hours after Sclerotinia sclerotiorum inoculation,reaching the maximum value at 12 hours.The results confirmed that BnRRS1-like and BnRRS2-like were disease resistant genes in B.napus.
